Rudi Hendriks did his PhD studies on X chromosome inactivation patterns in human X-linked immunodeficiency diseases in the Department of Immunohaematology (Prof. J.J. van Rood). After a post-doctoral training in the Genetics Laboratory of the University of Oxford, where he studied genetic aspects of X chromosome inactivation, he received a Royal Academy of Arts and Sciences fellowship and started his own line of research within the lab of Frank Grosveld at the Department of Cell Biology and Genetics of the Erasmus MC Rotterdam. In 1999 he moved his group to the Department of Immunology at the Erasmus MC, and, more recently, to the Department of Pulmonary Medicine.

His research focuses on the molecular biology of the developmental program of B and T lymphocytes. The largest effort has gone into the generation of transgenic and knock-out mice to investigate the role of Bruton's tyrosine kinase, the gene defective in the human immunodeficiency disease X-linked agammaglobulinemia. These studies identified novel functions for Btk in pre-B cells, as a regulator of immunoglobulin light chain rearrangements and as a tumor suppressor that limits pre-B cell expansion. The aims comprise the characterization of signal transduction pathways and lymphoid-specific transcription factors that implement cell fate decisions at specific checkpoints, controlled by antigen receptors and their immature forms.

The research laboratory of the Department of Pulmonary Medicine focuses on clinical and translational research concerning the pathology of various respiratory disorders. In this context, we study the molecular biology of the differentiation program of T helper subsets and to identify their role in the pathogenesis of various inflammatory diseases, including asthma, pneumonia and sarcoidosis. In addition, we study the role of dendritic cells in directing and maintaining immune responses in the lung, e.g. in influenza virus infection or in asthma. Hereby, we focus on signal transduction pathways downstream of Toll-like receptors in particular. Finally, we aim to develop dendritic-cell based immunotherapy in mesothelioma, a tumor caused by asbestos fibre inhalation.
